Griseofulvin, an antifungal medicine, is used to treat fungal infections in the nails or scalp. You can take it by mouth. Side effects include allergic reactions and nausea. diarrhea can also be caused by this drug. People with liver disease or porphyria should not use it. Griseofulvin interferes with fungal mitosis. It is a component of the WHO’s list of essential medications. Griseofulvin can only be taken orally for Dermatophytosis. Griseofulvin is not effective topically. This is for situations where topical creams are ineffective. For Microsporum scalp infections, griseofulvin is better than terbinafine. The drug binds tubulin and interferes with microtubule functions, thus interfering in mitosis. It is able to bind to Keratin in keratin-precursor cells, making them immune to fungal infections. Only after the keratin/griseofulvin complex is added to hair or skin, does the drug reach its intended site of action. Griseofulvin enters dermatology via energy-dependent transport processes and binds with fungal microtubules. This changes the process for mitosis as well as the underlying information to the deposition of fungal cell walls. P. is responsible for the first step in the biosynthesis process of griseofulvin.

The 14-carbon poly-b-keto chain undergoes cyclization/aromatization, using cyclase/aromatase, respectively, through a Claisen and aldol condensation to form the benzophenone intermediate. The methylated intermediate is then adenosyl methyl (SAM), twice to produce griseophenoneC. The halogenated species is then subject to a single phenolic oxygenation in each ring, forming two oxygen diradicals species. The right oxygen radical changes alpha to carbonyl via resonance, allowing for stereospecific radical coupling with the oxygen radical on the left ring. This forms a tetrahydroquinone type. To generate dehydrogriseofulvin, the newly formed grisan structure with a spiro center is O-methylated using SAM. Finally, griseofulvin is obtained by a stereoselective decrease of the olefin in dehydrogriseofulvin using NADPH.
